There's a clear difference:

 

"pendant une heure""for an hour"

 

Expressing for + [duration] in French with either pendant/durant/depuis/pour (French Prepositions of Time)

"en une heure""(with)in an hour" 

En vs Dans with time (French Prepositions of Time)
